World of Temperly will be located in a six storey Georgian building on Bruton Street and will also become the new head office.
The announcement has come some ten years after Somerset-born Alice launched her first collection at London Fashion Week.
Miss Temperley, whose fans include the Duchess of Cambridge, Sienna Miller, Eva Mendez and Emma Watson, was awarded an MBE for her services to fashion, last year.
She said that now felt like the "right time" to take her business to the next level, by opening the store.
The store will settle itself among some of the finest restaurants and shops in the city, and is set to open this November.
